The first successful underwater launched missile that could actually be counted on to hit its target was Polaris. Tests began in 1958, with the first successful launch in April 1959. This was from a test rig, not a submarine, as the first true missile sub, USS George Washington, was still incomplete. GW entered service and fired its first missile in 1960.
Previous American submarine launched missile tests were with cruise missiles, requiring surfacing, setup, and ramp launches. These started with the Loon missile, which was an American-made version of the German V-1. Regulus followed, but the advent of the SLBM pretty much ended American interest in cruise missiles until the development of modern types that can be fired from a torpedo tube without surfacing.
